## Mechanism

GHK-Cu works by delivering copper to tissue, enabling the collagen- and elastin-cross-linking enzyme lysyl oxidase, and by acting as a broad gene-expression signal that tilts cells toward repair and away from inflammation [2]. GLOW layers two additional mechanisms on top of GHK-Cu's: BPC-157's pro-angiogenic, VEGFR2-driven vascular protection [10], and TB-500's actin-binding, cell-migration effects — three distinct mechanisms bundled into one product, never tested together. Melanotan II works through an entirely different system: non-selective activation of melanocortin receptors, which raises pigment production through MC1R and separately affects appetite and sexual response through MC3R/MC4R in the brain. The three compounds do not compete for the same biological territory — they sit in genuinely different parts of the skin-aging and research-peptide landscape.

## Evidence maturity

GHK-Cu is, by a wide margin, the most mature of the three: decades of cell and animal work, a real body of small human topical trials, and one placebo-controlled human hair-growth trial with a statistically significant result [3]. GLOW inherits none of that maturity as a combination — every specific claim on its page traces to a single-ingredient study, and the 2026 review that names all three ingredients together is explicit that rigorous human safety data for compounds like these remain scarce [8]. Melanotan II's efficacy evidence is real but small in scale — a 10-person crossover trial for its erectile effect [15] — while its safety literature is unusually extensive for a compound this size, almost entirely because of published case reports documenting serious harm.

## Safety profile

This is where the three separate most sharply. GHK-Cu's safety concerns are mostly about topical irritation and formulation compatibility, with theoretical (not documented) concerns about systemic copper accumulation if it were used by injection. GLOW inherits GHK-Cu's copper caution plus two additional and more serious ones: its TB-500 ingredient is WADA-prohibited for tested athletes, and its BPC-157 ingredient's pro-angiogenic mechanism raises a theoretical caution for anyone with an active or recent cancer. Melanotan II carries the heaviest documented burden of the three — published case reports of new and changing moles, melanoma, rhabdomyolysis, kidney infarction, priapism, and a brain-swelling condition (PRES), on top of a market where analytical testing repeatedly finds mislabeled or inconsistent product. None of the three is approved for any anti-aging or cosmetic indication in the way this site discusses them, and none should be read as recommended for human use here.
